3.2 Step-by-Step Usage Process


1. Tissue Infiltration: After fixation, dehydration, and clearing, place the tissue samples in a tissue processor with melted paraffin wax (heated to 2-3℃ above its melting point). The high permeability of our wax ensures full infiltration into the tissue, replacing all clearing agent.


2. Embedding: Pre-warm the embedding mold, pour a small amount of melted wax into the bottom, place the oriented tissue sample in the mold, and fill the mold with wax. Attach the labeled cassette to the mold and place it on a cold plate to solidify.


3. Block Trimming: Once the wax block is fully solidified, remove it from the mold and trim the excess wax around the tissue, creating a smooth, trapezoidal surface for slicing.


4. Sectioning: Mount the wax block on a rotary microtome, adjust the slicing thickness (typically 4-5μm for routine H&E staining, 2μm for special staining), and cut continuous sections. The pearl-shaped wax ensures smooth, wrinkle-free sections.


5. Flotation & Mounting: Float the sections on a 45℃ water bath to remove wrinkles, then mount them on glass slides and dry on a slide warmer before staining.
